The mRNA vaccines are currently being researched for applications in various diseases by delivering mRNA into cells. To deliver mRNA into cells, it’s important to protect it from extracellular elements like nucleases. This can be achieved by encapsulating mRNA with polymers to form polymer nanoparticles (PNPs), enabling delivering mRNA into cells. Poly-β-peptide derivatives were synthesized with various degree of polymerization (DP) while maintaining high yields and low polydispersity by using β-amino acid N-thiocarboxyanhydrides (β-NTAs) via open-vessel ring-opening polymerization. Additionally, PGly(DET/CHE) were developed, incorporating diethylenetriamine (DET) for adjusting pKa values to facilitate endosomal escape and 2-cyclohexylethylamine (CHE) for regulating log P values to control colloidal stability. As a result, the physiological and biological properties of PNPs were analyzed based on PEG length and further assessed mRNA expression levels in cells and mice. |
